I started this new opportunity a week ago, but it's not right for me. I feel awful relating my manager because she be training me to be assistant manager.

you'll inevitability to check your employment contract on this one. some temping agencies give you pro rata holiday (e.g. if your contract say you get 20 days holiday a year, you'll receive that proportionally for every week you work so that if you worked a full year, you'd own 20 days holiday owing to you).
some of the time they do it so that you can't take more than a few days stale at a time, or you can't 'roll over' one month's holiday to the next. again, it will be within your employment contract. if you don't have a copy of your contract, jump to your agency and ask for one. you have a legitimate right to have it.
so long as your boss at the workplace you're workign at allows you to clutch that holiday, you can take it. if the agency hires someone to give somebody a lift your place while you're gone, they will be in breach of contract. similarly, if you are refuse the (weekly or monthly) equivalent of 20 days per year of paid holiday, they are breaking the ruling in the UK.
if you've get any complex queries, it's best to combine a union to protect yourself. agencies can do dodgy stuff and try to return with away with it. (my coalition is the IWW. they're small, but very pious at what they do). remember, you're techincally being employed by the agency, not your boss at your workplace - no situation what they say to the contrary!
