I would suggest hiring a web designer but if you want to go the quick (but not necessarily easier or better) route, you should do the following:
1. Get a domain (I recommend skipping this and going to number 2)
2. Get hosting...I use 1and1 which includes free domain registration for their packaging. If you plan on doing it yourself, I would get hosting that has a website builder integrated into it.
3. Build the site
4. Upload it
Without any more background it's hard to say what you are wanting to do. For e-commerce I like Mals as it's free to set up and also free to run unless you need more advanced features.

First what type of business are you starting? Are you selling items, offering services...etc?
If you pay sale tax on the item, you enjoy to charge the buyer sales charge. (That is how it is here in Tennessee anyway). You call for to contact your State and find out exactly what you must do. When my husband and I started our business, I got on a first moniker business with our State business dept and the IRS. Don't be afraid to ring up and ask questions, its better to be sure, later to mess up and have to jump through all the crap to correct it.
